Learning Outcomes and Application This unit of competency describes the skills and knowledge to perform aseptic techniques to maintain the integrity of both the sample source and the sample. It applies to sampling techniques in tissue culture and to generic microbiological procedures.This unit of competency applies to laboratory assistants and technicians working in the field or laboratory in the biomedical, biological, food processing and environmental industry sectors.No licensing or certification requirements exist at the time of publication. However, regulations and/or external accreditation requirements for laboratory operations exist, so local requirements should be checked. Relevant legislation, industry standards and codes of practice within Australia must also be applied.
Duration and Setting X weeks, nominally xx hours, delivered in a classroom/online/blended learning setting.

Elements of Competency Performance Criteria              
Element: Prepare for aseptic sampling or transfer
  • Check the sampling procedure conforms with the required sampling plan
       
Element: Use specified personal protective equipment (PPE)
       
Element: Prepare the work area for safe and effective sample transfer
       
Element: Select equipment and materials specified by the procedure
       
Element: Organise equipment to minimise contamination during manipulations
       
Element: Label containers for clear identification
       
Element: Record details in relevant log or database
       
Element: Macroscopically assess media and/or reagents to confirm sterility before use
       
Element: Transfer materials aseptically
  • Protect the integrity of the sample source
       
Element: Sterilise inoculating loops and/or pipette where used to prevent contamination
       
Element: Perform transfer while minimising opportunities for contamination and cross-infection
       
Element: Protect the integrity of the sample source and destination after transfer, and before sealing the transport or culture vessel
       
Element: Re-sterilise inoculating loops, minimising the generation of aerosols as required
       
Element: Perform quality control checks to confirm aseptic transfer was successful
       
Element: Label transport or culture vessels for clear identification
       
Element: Maintain work area and equipment to prevent cross-infection and contamination
  • Place disposable and reusable items into relevant receptacles
       
Element: Clean and disinfect work area and equipment after use
       
Element: Transport disposable and reusable contaminated materials to relevant areas for disinfection, sterilisation and cleaning or disposal
       
Element: Follow personal hygiene procedures
